Azides:

Several of our specialty dyes are now available as azides to be reacted with C≡C-triple bonds in either a Cu(I)-catalized or Cu-free 1,3-dipolar cycloaddition reaction to a triazole (see figure). Both reactions proceed in aqueous environments with adequate efficiency.

SeTau-405-azide is a clickable dye with a longer lifetime that is well suited for excitation with the blue, 404 nm diode laser. It has a large Stokes’ shift of over 110 nm, and the 8 ns lifetime make it also a valuable label for studying protein hydrodynamics (fluorescence polarization measurements of higher MW analytes). It is a good alternative for Pacific Orange to be used with Seta-405 in flow cytometry applications. Compatible with Fluorescein emission filter. Exc./Em. max: 405/518 nm.

The lifetime-sensitive Seta labels Seta-633-azide or Seta-670-azide exhibit shorter fluorescence lifetimes (FLTs) in aqueous media or when labeled to low molecular weight analytes (drugs or small ligands) but increased brightness and up to ten-times longer FLTs when bound to proteins or oligonucleotides and are unique tracers for FLT-based assays.

Seta-670-azide is also a label with extremly high 2PACS of 2000 GM at 840 nm for use in 2-photon microscopy.

 

Cyclo-Octyne (DBCO) Derivatives:

Strain-mediated cyclization reactions of cyclo-octynes proceed without the need for a Cu-catalyst. The metal-free reaction has wider applicability and is compatible with cell-based applications.

SeTau-647-DBCO is an extremely bright, hydrophilic click chemistry reagent containing one dibenzylcyclooctyne (DBCO) group for strain-mediated cycloaddition reactions with azides. It has the same excitation wavelength as Cy5, Alexa Fluor™ 647 and can therefore be used with these filter sets. Its emission is at 690 nm.
 
Other available DBCO-modyfied dyes are Seta-555-DBCO, Seta-650-DBCO and Seta-750-DBCO.
